Histo- and Cytological Characteristics of Organogenic and Non-organogenic Callus in Gladiolus ¢¥ Topaz ¢¥



Abstract
This study was performed to compare characteristics between organogenic callus (OGC) and non-organogenic callus (NOGC) of Gladiolus ¢¥Topaz¢¥, which were subcultured on MS solid medium with 0.5 §·¡¤L^(-1) 2,4-D. NOGC showed characteristic of the typical mature or senescence tissue in comparison with OGC. That is, NOGC had the fewer number of cells per fresh weight and bigger cells than OGC and the vacuole and the secondary cell wall of NOGC were well developed in comparison with OGC. In NOGC, cell organelles were also abnormal and endogenous ethylene were released more than OGC. Thus, development of abnormal cell organelles in NOGC was presumed that 2,4-D in medium caused the change of pH in membrane. Peroxidase was localized mainly in the cell wall of surface in OGC.
